It would be better to take a train to go to Florence since Florence is situated on the main line between Milan and Napoli.
Every train like Eurostar and other local trains stops at the SMN station in Florence.
If you go to Florence from Rome, it could be fine to stop over in Orvieto or Arezzo: they are also marvelous cities.
If you want to go to other beautiful towns in Tuscany, Florence would be a hub spot.
You can take a direct bus from Florence to; Siena, a town where there is Piazza del Campo, the most magnificent square in the world, San Gimignano, a town where there are plenty of beautiful towers and Lucca, a town where Puccini was born.
3-5 trains are available per an hour between Rome and Florence including Eurostar, Intercity and local express trains.

You can get to Siena or San Gimignano from Florence by the bus which is operated by SITA or TRA.IN.
The terminus of them is located at the west side of SMN station, the terminal of SITA, Stazione dell’autobus. You can refer to the detailed timetable information inside this terminal: tickets are also available inside.
If you want to go to Siena or San Gimignano, you will enjoy by the one-day trip; it would be convenient to buy a round trip ticket in Florence and it would be better to check the timetable for your return.
You are required to punch your ticket when you get on the bus, like when you take a train; if you don’t have a valid ticket, you will be fined. Also inside of the terminal you can find a validation machine. Don’t forget to punch before riding.
To get to Siena, take the bus operated by SITA or TRA.IN.
The terminus is located at the west side of SMN station, the terminal of SITA. One-way ticket costs 6.50 euro.
It take 70 minutes by “Siena Rapida” (non-stop bus to Siena), and take 90 minutes by “Siena Diretta” (stops also in other towns, a local bus).
